<p><tp:taxon-name><tp:taxon-name-part taxon-name-part-type="genus" reg="Eucorydia">Eucorydia</tp:taxon-name-part></tp:taxon-name> Hebard, 1929 exhibits striking metallic coloration and bright markings, making it one of the most morphologically and aesthetically distinct lineages within <tp:taxon-name><tp:taxon-name-part taxon-name-part-type="superfamily" reg="Corydioidea">Corydioidea</tp:taxon-name-part></tp:taxon-name>. The genus is widely distributed across East Asia, Southeast Asia, and South Asia, with 22 known species worldwide, ten of which occur in China (<xref ref-type="bibr" rid="B1">Asahina 1971</xref>; <xref ref-type="bibr" rid="B35">Woo et al. 1986</xref>; <xref ref-type="bibr" rid="B25">Qiu et al. 2017</xref>; <xref ref-type="bibr" rid="B36">Yanagisawa et al. 2021a</xref>).</p>
<p>Early taxonomic studies relied primarily on external morphological characters, such as body color, tegminal and abdominal markings (<xref ref-type="bibr" rid="B23">Princis 1963</xref>; <xref ref-type="bibr" rid="B1">Asahina 1971</xref>; <xref ref-type="bibr" rid="B35">Woo et al. 1986</xref>; <xref ref-type="bibr" rid="B34">Woo and Feng 1988</xref>). Subsequent revisions incorporated male genitalia, greatly improving species delimitation (<xref ref-type="bibr" rid="B25">Qiu et al. 2017</xref>, <xref ref-type="bibr" rid="B26">2019</xref>). More recently, molecular approaches, including genetic distance analyses and phylogenetic inference, have further enhanced taxonomic resolution within the genus (<xref ref-type="bibr" rid="B36">Yanagisawa et al. 2021a</xref>, <xref ref-type="bibr" rid="B37">2021b</xref>, <xref ref-type="bibr" rid="B38">2021c</xref>). Despite these advances, studies of <tp:taxon-name><tp:taxon-name-part taxon-name-part-type="genus" reg="Eucorydia">Eucorydia</tp:taxon-name-part></tp:taxon-name> remain strongly biased toward males. Females have received little attention, with most studies limited to simple descriptions and comparisons of external morphology (<xref ref-type="bibr" rid="B25">Qiu et al. 2017</xref>). However, previous research has shown that the sclerotized structures of female genitalia in <tp:taxon-name><tp:taxon-name-part taxon-name-part-type="order" reg="Blattodea">Blattodea</tp:taxon-name-part></tp:taxon-name> are stable and species-specific, serving as important diagnostic characters (<xref ref-type="bibr" rid="B20">McKittrick 1964</xref>; <xref ref-type="bibr" rid="B39">Zhu et al. 2022</xref>; <xref ref-type="bibr" rid="B31">Valero-Pérez 2023</xref>; <xref ref-type="bibr" rid="B10">Han et al. 2024</xref>). Such characters have not yet been systematically investigated in <tp:taxon-name><tp:taxon-name-part taxon-name-part-type="genus" reg="Eucorydia">Eucorydia</tp:taxon-name-part></tp:taxon-name>. Furthermore, molecular data for Chinese species remain limited, despite China harboring nearly half of the known diversity of the genus.</p>
<p>Cytochrome c oxidase subunit I (<abbrev xlink:title="Cytochrome c oxidase subunit I">COI</abbrev>), especially the standard 658-bp barcode fragment, is widely recognized as a rapid and reliable molecular marker for species identification (<xref ref-type="bibr" rid="B12">Hebert et al. 2003</xref>; <xref ref-type="bibr" rid="B18">Liao et al. 2019</xref>). It has been successfully applied to insect species delimitation using the Assemble Species by Automatic Partitioning (<abbrev xlink:title="Assemble Species by Automatic Partitioning">ASAP</abbrev>) and Generalized Mixed Yule Coalescent (<abbrev xlink:title="Generalized Mixed Yule Coalescent">GMYC</abbrev>) methods (<xref ref-type="bibr" rid="B2">Bai et al. 2018</xref>; <xref ref-type="bibr" rid="B24">Puillandre et al. 2021</xref>; <xref ref-type="bibr" rid="B21">Neo et al. 2025</xref>).</p>
<p>In this study, we re-evaluate the species diversity of <tp:taxon-name><tp:taxon-name-part taxon-name-part-type="genus" reg="Eucorydia">Eucorydia</tp:taxon-name-part></tp:taxon-name> in China using an integrative approach combining morphology and molecular data. Furthermore, we provide the first comparative analysis of female genitalia in this genus and evaluate their taxonomic significance.</p>

<p>For phylogenetic analysis, the optimal partition scheme and substitution model were determined using PartitionFinder v. 2.1.1 (<xref ref-type="bibr" rid="B17">Lanfear et al. 2017</xref>) with default parameters. The maximum likelihood (<abbrev xlink:title="maximum likelihood">ML</abbrev>) phylogenetic tree was reconstructed with ten independent likelihood searches performed in IQ-TREE v. 2.2.0 (<xref ref-type="bibr" rid="B22">Nguyen et al. 2015</xref>) to identify the best-supported topology. The result with the highest likelihood value was selected for presentation. Node and branch support was evaluated with 10,000 ultrafast bootstrap (<abbrev xlink:title="ultrafast bootstrap">UFBoot</abbrev>) replicates. In addition, the ‘-bnni’ option was used to mitigate severe model violations, and finally visualization was performed using FigTree v. 1.4.4 (<ext-link xlink:href="http://tree.bio.ed.ac.uk/software/figtree/" ext-link-type="uri">http://tree.bio.ed.ac.uk/software/figtree/</ext-link>). Molecular species delimitation was performed using the <abbrev xlink:title="Assemble Species by Automatic Partitioning">ASAP</abbrev> method (<xref ref-type="bibr" rid="B24">Puillandre et al. 2021</xref>), based on pairwise <abbrev xlink:title="Kimura 2-parameter">K2P</abbrev> genetic distances with default parameters and a recursive split threshold of 0.01. The optimal species partition was determined by the minimum <abbrev xlink:title="Assemble Species by Automatic Partitioning">ASAP</abbrev> score, which integrates the probability of panmixia and the relative barcode gap width.</p>
<p>The <abbrev xlink:title="Generalized Mixed Yule Coalescent">GMYC</abbrev> method requires a fully resolved, strictly ultrametric bifurcating tree as input. To meet this requirement, a Bayesian inference tree was reconstructed in BEAST 1.8.2 using the best-fit nucleotide substitution model selected by PartitionFinder2. An uncorrelated lognormal relaxed-clock model was applied to accommodate rate variation among branches. The mean clock rate was estimated under a uniform prior distribution with a lower boundary of 0, an upper boundary of 1, and an initial value of 0.0001. A UPGMA tree was used as the starting tree, and a birth-death model was specified as the tree prior. Two independent Markov Chain Monte Carlo (<abbrev xlink:title="Markov Chain Monte Carlo">MCMC</abbrev>) analyses were executed for 5 million generations each, with sampling every 500 generations. The resulting trees were then summarized using TreeAnnotator in the BEAST package to generate a maximum clade credibility tree, with the first 10% of sampled trees from each run discarded as burn-in. Convergence was assessed with effective sample size (<abbrev xlink:title="effective sample size">ESS</abbrev>) values ≥ 200 in Tracer v. 1.6.0 (<xref ref-type="bibr" rid="B3">Drummond and Rambaut 2007</xref>).</p>
<p>The final ultrametric gene tree was submitted to the <abbrev xlink:title="Generalized Mixed Yule Coalescent">GMYC</abbrev> online server (<ext-link xlink:href="https://species.h-its.org/gmyc/" ext-link-type="uri">https://species.h-its.org/gmyc/</ext-link>) for species delimitation using the single-threshold algorithm. The backend of this web server implements the original R version of the <abbrev xlink:title="Generalized Mixed Yule Coalescent">GMYC</abbrev> model (<xref ref-type="bibr" rid="B5">Fujisawa and Barraclough 2013</xref>). The delimitation results were evaluated using a likelihood-ratio test, which statistically compares the multiple-species clustering model with a single-species null model.</p>

Comparative analysis reveals consistently high sclerotization in four key structures across all examined species: spermathecal plate, basivalvula, vestibular sclerite, and laterosternite IX, which also exhibits species-specific variation, supporting their taxonomic value. The sclerotized portions of the basivalvular lobe are relatively narrow and are connected to the spermathecal plate by a membranous structure (Figs <xref ref-type="fig" rid="F4">4E</xref>, <xref ref-type="fig" rid="F6">6</xref>). The crosspiece typically articulates with the first valvifer and laterosternite IX, extending toward the posterior lobe of valvifer II (Figs <xref ref-type="fig" rid="F4">4E</xref>, <xref ref-type="fig" rid="F6">6</xref>), which exhibits interspecific variation in both shape and size. <p>In this study, the minimum interspecific <abbrev xlink:title="Cytochrome c oxidase subunit I">COI</abbrev> genetic distance was found to be 3.39%, observed between the <tp:taxon-name><tp:taxon-name-part taxon-name-part-type="genus" reg="Corydia"/><tp:taxon-name-part taxon-name-part-type="species">zonata</tp:taxon-name-part></tp:taxon-name> population of <tp:taxon-name><tp:taxon-name-part taxon-name-part-type="genus" reg="Eucorydia">E.</tp:taxon-name-part> <tp:taxon-name-part taxon-name-part-type="species" reg="dasytoides">dasytoides</tp:taxon-name-part></tp:taxon-name> and <tp:taxon-name><tp:taxon-name-part taxon-name-part-type="genus" reg="Eucorydia">E.</tp:taxon-name-part> <tp:taxon-name-part taxon-name-part-type="species" reg="qinlingensis">qinlingensis</tp:taxon-name-part></tp:taxon-name> sp. nov. This value falls well below the maximum intraspecific variation of 8.67%, which was detected between the geographically separated samples from Guizhou and Taiwan Provinces, thereby resulting in an overlap in the DNA barcoding gap. Consequently, the distance-based <abbrev xlink:title="Assemble Species by Automatic Partitioning">ASAP</abbrev> analysis fails to delimit the two lineages as separate <abbrev xlink:title="molecular operational taxonomic units">MOTUs</abbrev>. Nevertheless, morphological evidence, particularly the shape of the posterior margin of the male supra-anal plate, combined with results from the tree-based <abbrev xlink:title="Generalized Mixed Yule Coalescent">GMYC</abbrev> analysis, supports their recognition as distinct species.</p>
<p>Compared with related corydiid cockroaches, the minimum interspecific distance detected in <tp:taxon-name><tp:taxon-name-part taxon-name-part-type="genus" reg="Eucorydia">Eucorydia</tp:taxon-name-part></tp:taxon-name> is relatively low. For example, the reported minimum interspecific <abbrev xlink:title="Cytochrome c oxidase subunit I">COI</abbrev> distances are 9.8% in <tp:taxon-name><tp:taxon-name-part taxon-name-part-type="genus" reg="Eupolyphaga">Eupolyphaga</tp:taxon-name-part></tp:taxon-name> and 5.80% in <tp:taxon-name><tp:taxon-name-part taxon-name-part-type="genus" reg="Pseudoeupolyphaga">Pseudoeupolyphaga</tp:taxon-name-part></tp:taxon-name> (<xref ref-type="bibr" rid="B9">Han et al. 2022</xref>; <xref ref-type="bibr" rid="B27">Ren et al. 2025</xref>). However, an overlap between intra- and interspecific genetic distances has also been reported in these genera, with maximum intraspecific distances reaching 9.54% in <tp:taxon-name><tp:taxon-name-part taxon-name-part-type="genus" reg="Eupolyphaga">Eupolyphaga</tp:taxon-name-part></tp:taxon-name> and 6.90% in <tp:taxon-name><tp:taxon-name-part taxon-name-part-type="genus" reg="Pseudoeupolyphaga">Pseudoeupolyphaga</tp:taxon-name-part></tp:taxon-name> (<xref ref-type="bibr" rid="B9">Han et al. 2022</xref>; <xref ref-type="bibr" rid="B27">Ren et al. 2025</xref>). These comparisons suggest that the absence of a clear DNA barcoding gap may not be unique to <tp:taxon-name><tp:taxon-name-part taxon-name-part-type="genus" reg="Eucorydia">Eucorydia</tp:taxon-name-part></tp:taxon-name>, but may occur more broadly among related corydiid taxa.</p>
<p>These results highlight the limitations of relying exclusively on <abbrev xlink:title="Cytochrome c oxidase subunit I">COI</abbrev> sequences for species delimitation in <tp:taxon-name><tp:taxon-name-part taxon-name-part-type="genus" reg="Eucorydia">Eucorydia</tp:taxon-name-part></tp:taxon-name> and related genera, as single mitochondrial markers may be affected by incomplete lineage sorting, introgression, and recent divergence (<xref ref-type="bibr" rid="B7">Funk and Omland 2003</xref>; <xref ref-type="bibr" rid="B4">Dupuis et al. 2012</xref>). Although external morphology and color pattern are useful for preliminary identification, they may be insufficient when closely related species show limited mitochondrial divergence or when geographically structured populations exhibit high intraspecific variation (<xref ref-type="bibr" rid="B33">Will et al. 2005</xref>). Therefore, future studies should integrate broader geographic sampling, quantitative morphometric analyses, multi-locus nuclear markers, and genomic-scale data to clarify species boundaries and evaluate cryptic diversity in corydiid cockroaches.</p>
